Swing Valley Poem Analysis

Pursue of happiness Contentment can help people move forward, it is also a way of freezing time to connect to the past. To compromise to happiness is hardly possible. However in the poem Swing Valley, and the excerpt from Home Place, the speakers are able to find joy by means of nostalgia. They seem to pursue happiness by recollecting favourable memories from the past, and being in the physical reminiscence that shapes their past. Through the development of the character, Ronald, and the speaker from Swing valley, we are able to apprehend how recollection memories can lead to happiness. When it comes to ecstasy, it’s not a word in Ronald’s dictionary. Though, as he revisits his child-hood memories, everything suddenly becomes clear. As Ronald was looking for a companionship in life, he jumped from the frying pan and into the fire. â€Å"’I can’t stand it. I can’t sleep there no longer. ’† The regret and sorrow he’s feeling made h im longing for joy. Through further understanding of Ronald, we realise that he is feeling nostalgic towards his past, his childhood, and his true passion – creating model airplanes. Primarily, he pursued for happiness as he married Darlene, out of lust, and loneliness. For what seemed to Ronald was unattainable, he later discovers that joy was something that actually could be reached by recalling what he loved, rather than what he thought he loved. He found himself under the silhouettes of his model air planes that covered his walls. He found himself understanding the concept of delight. He found himself having minimal interest in anything other than the model air planes that he loved even as a young man. Simple moments such as gravitational loss can help us discover happiness in the present. Ordinary may have different meanings to different individuals. Some ordinary flashes of nostalgia have helped people renew connections of the past to the present. The descriptiveness of F rank Gaspar’s Swing Valley shows the readers how much a nostalgic event can sink so deeply to one’s memory. Even the precise description of â€Å"still sitting on the knot, his hands clutching the rope as the bitter end trailed down like a plume of smoke. † The speaker has strong feelings of the tense atmosphere that they were in, as he relates it to the moment of release. In life, this is a metaphor of that moment of finally achieving the goal you’ve trying to accomplish for years. As the speaker recalls that moment, he immediately thinks of the time when he’s free of gravity. It wasn’t just a rope swing. As Ronald finds joy, he wishes that he â€Å"should never have left. † Regretting how much happier life should have been. The speaker from Swing Valley finds happiness through remembering the wonderful memories of the past and reflects the present with it. These characters discovers the pursuit of happiness as they come across moment s of nostalgia, and as they’re longing for their past to compromise their happiness in the present.

The Little Match Girl

THE LITTLE MATCH GIRL -Hans Christian Andersen- Hans Christian Andersen (1805-1875) is a Danish author, fairy tale writer, and poet noted for his children’s stories. Although during his lifetime he was acclaimed for having delighted children worldwide, his stories were always meant for adults as well. His poetry and stories have been translated into more than 150 languages. They have inspired motion pictures, plays, ballets and animated films. However, fairy tales take the most important role in his writing career. Andersen used the colloquial style that disguises the sophisticated moral teachings in his fairy tales. Therefore, his tales became the favorite ones of both children and adults. Some of the most famous tales of Andersen are â€Å"The Little Mermaid†, â€Å"The Ugly Duckling† and â€Å"The Little Match Girl†, etc. Especially, â€Å"The Little Match Girl† which is famous for its humane quality has shaken the hearts of millions of readers in the world. The story is firstly published in 1845. This one begins with a little girl who appears to be alone and very cold at New Year’s Eve. She is a poor girl who tries to sell matches at the cold night. She doesn’t come back home because she is very afraid of her father who let her out to sell matches. After finding a place between two houses’ wall, the poor girl starts to light matches to warm herself. When matches are burning, she can see many things which make her very happy. Such as: a Christmas tree, a holiday feast, a shooting star, etc. Then, the little girl sees a vision of her grandmother, the only person to have treated her with love and kindness. Therefore, she lights other matches to keep her grandmother stay nearby as long as she can. Finally, she can go together with her grandmother to the heaven. The next morning, people find her dead on the snow but there is still the smile on her red cheek. In the story, there are two significant literature elements are character and the symbol. They are the most important factors that make the story become so successful. Firstly, the story has only one main character: the poor little match girl. Andersen didn’t give her a specific name in order to emphasize her hard life. Through the way writer introduce the main character, readers can see that the little girl is also one among many poor children like her. In fact, there are also three other characters. They are her family members: grandmother, dad and mom. However, all of them are indirectly described through the girl’s appearance, thought and imagination. At the beginning of the story, she appears to be bareheaded and barefoot in a terribly cold night. She is sent out to sell matches in the snow with a pair of ill-fitting slippers which used to belong to her mother. However then she loses one when she is running across the road and when a boy take up the rest. The father appears in her thought as a very irresponsible father. Although she is very hungry and cold, she doesn’t dare to go home. With just a line of thought, a father who doesn’t love his daughter is introduced clearly. Finally, her last family member, her grandmother, is mentioned affectionately through her short time life of each burning match. In the story, there are nothing related to the mother except for her ill-fitting slippers. However, the little match girl have no memories about her mother. While she fears the most is her father, grandmother is the one she does love who died for such a long time ago. The way Andersen indirectly introduces about these three characters highlights the loneliness and the unhappiness of the poor little girl. There is no one in the world that cares for her, they just try to step as quickly as they can to avoid the cold, late night without noticing of a lonely poor girl. Secondly, the story has a significant symbol which is always in readers’ mind. It is the light of match. After each time the poor girl strikes match against the wall, the match burns brightly and she is immersed in  the illusion  of happiness. Firstly she imagines that she was sitting before a great iron stove. The next time, the girl sees a table with spread snow-white cloth and a lot of delicious food prepared for the dinner. Then, the most beautiful Christmas tree seems to be right beside her. After that, she meets her beloved grandmother and finally, she continuously strikes the whole bundle of matches in order to keep the grandmother stay with her. After each time the flame go out, everything is vanished rapidly. Therefore, she has to come back to the hard reality. It can be said that the light in this story is the symbol of dreams of happiness. Everything she needs at the present becomes clearer than ever in the light of the match flame. She just dreams of many things which are normal to almost everyone but very luxury to her. The light is the bridge of her hard reality and the happy dreams. Therefore, the appearance of the light throughout the story highlights the emotions, wishes and dreams of the poor little girl. She always dreams of the good things even though the life has treated her so hard. Is the light also the wish of the author to the little girl and all of other poor people? Through the little girl’s dream, he expresses his mercy to these unfortunate. Using metaphor device through the light image, Andersen successfully describes the dreams of happiness, the little girl’s short life and even the humane value message he want to send to readers. This symbol greatly contributes to the vitality and humane quality of the story. To sum up, the story â€Å"The Little Match Girl† has the great reality and humane value. It indirectly reflects tragic fate of the poor in the current Danish society. Moreover, through the story we can see clearly the compassionate heart of the author and his talent as well. Norman Rockwells Painting Essay Example | Topics and Well Written Essays - 500 words

Norman Rockwells Painting - Essay Example This article states that the painting was inspired by Rockwell’s when he visited the Soviet Union. The painting offers a learning experience for teachers, students, and adult learners. The artist had an insightful observation of Russian classroom experience of schoolchildren in the 1960s. The artist was reflecting what he observed during his visit and was amused with what he saw. This made him to reflect but and inspire other students in both upper and lower primary. It was also an inspiration to parents and teachers on the extent to which children can stay focused on their own. The author believes that the problem we all live with has been embraced and become an important symbol of civil rights. It acts as a remembrance of the day when the United States Supreme Court declared the racial segregation unconstitutional. The painting bears a black girl who is determined to go to school and she is under escort. Additionally, the author asserts that the painting is a symbol of victo ry on the black race following the court verdict that state law establishing separate public schools for whites and blacks were unconstitutional. The artist was quick to take note because he was a supporter of tolerance and equality. The author outlines that the federal Supreme Court declared segregation illegal in public schools. Norman Rockwell drew the picture to show a young black girl in a uniform walking to school in little rock. The child is protected by US federal marshals, something which has never been realised.

Investigating Decision-Making Methods Essay Example | Topics and Well Written Essays - 1250 words

Investigating Decision-Making Methods - Essay Example Several decision-making strategies exist for such leaders and administrators. In considering which decision, making strategy to use at any particular time requires them to consider the specific public directly affected by the decision and the possible ramifications resulting from the target public’s reaction. As a school leader, Mr. Miller has several decision making strategies to consider o ensure that he makes relevant decisions that do not only safeguard the interests of the different communities but are ethical and perpetuate the moral development of the society at large. Among the strategies are simple prioritizing, satisficing and the pros and cons method. The simple prioritizing method dictates that a leader prioritizes effective by considering the pressing issues at a time. This way, the decisions they make redress the issues effectively thus providing them an opportunity to progress to other issues. Similarly, satisficing just like prioritizing demands that a leader c onsiders the alternatives presented by a scenario until they find the most appropriate. In determining the most appropriate alternative, one considers such factors as the moral and financial implications of the decision. Pros and cons just as the name suggests is a method that requires that leaders considers the positive and negative effects of the available alternative before taking an action. The method commands that the leader carries out adequate research and consultation to determine the effects of the decisions. Mr. Miller as a leader is likely to affect several people with the decisions he makes. It is therefore important that he find out the effects of such beforehand. It is only after finding out the probable effects that he makes a decision by ignoring the alternative that presents him with numerous negativities and selects an alternative with numerous positive effects. While the methods may sound numerous, they have similar structures and are effective in a democratic soc iety (Hill & Jones, 2011). As a school head, Mr. Miler must prove to the society that he is effective leader capable of making effectual decision and handle pressure. More importantly, Mr. Miller must take responsibility of the decision he makes. Taking responsibility is often one of the most difficult aspects of decision-making since some of such would include the negative outcomes of the decision like deteriorating performance or the misappropriation of the school funds. The three decision making strategies discussed above mitigate on the negativity of the outcome of the decision making process as the different publics contribute to the particular decision. Mr. Miller has managed to incorporate the community and different stakeholders to the school management by making inclusive decision. To sustain the relationship he has built with the different publics he must often consider his personality. Leadership relies on the personality, the subject rely on their leader for the moral an d psychological support. In the school setup, the rest of the staff and the collaborators would continue support the school despite its performance and outcome of the decisions only if Mr. Miller proves to be independent minded and inspiring. The good decision-making habits that Mr. Miller must possess include making decision under uncertainties. Different conditions present diverse challenges to the school ma

Terrorism Research Paper Example | Topics and Well Written Essays - 1250 words - 1

Terrorism - Research Paper Example Terrorism has existed throughout history and across different political systems. Just as there are many ways terrorism is carried out, many definitions of the term exist. However, all kinds of terror attacks revolve around use of violent methods of execution, targeting governments and civilians, with the sole purpose of inculcating fear to force social and political changes. Terrorism began way back in the century of Judea, and it has changed in organization, financing and tactics throughout the years. For example, with the emergence of technology, terrorist groups in the twenty first century have greatly utilized this technology to make their demands. Terrorist groups evolve and adapt according to constantly changing law enforcement tactics and intelligence agencies. This is evident when we look at present day activities of these groups and the fact that some even hide behind the veil of religious groups and organizations. All states and nations need to take necessary measures to pr otect the rights of every individual within their authority from terrorist attacks. Flexible counterterrorist policies and strategies need to be taken in order to be able to respond to the threat of terrorism, which is divided and wide in scope. Keywords: Terrorism, Terrorists, Terror Attacks, Organization, Financing, Tactics, Technology, Law Enforcement Tactics, Intelligence Agencies, States, Nations, Measures, Counterterrorist Policies Different forms of terrorism have been practiced all through history and across different political systems. ... In seventh century India, thugee cult members ceremonially throttled their victims in a practice of offering sacrifice to the Hindu goddess known as Kali (Kushner, 2003). The term terrorism appeared during the French revolution. The term was used by Edmund Burke, a British Political Philosopher, to define the situation in revolutionary Paris. White asserts that â€Å"He referred to the violence as the Reign of Terror, and he used the word terrorism to describe the actions of the new government† (2009, p.123). During the wars that took place in Napoleons reign, the meaning of terrorism begun to undergo slight transformations. The Spanish during the Peninsula War called partisans patriots, while the French called them terrorists. Thus â€Å"the meaning of terrorism shifted away from governmental repression and seemed to apply to those who resisted governments† (White, 2009, p.123). The transformation in the definition of what terrorism is continued into the nineteenth cen tury. The nature of European violence in the 1800s changed the meaning of terrorism in the minds of western people. Transition in the history of western terrorism was spearheaded by Heinzen and Most. Western terrorism had been formed by a circle of radical revolutionaries who administered mass executions on behalf of the French government. As Spanish partisans terrorized French troops with unconventional methods after the 1807 French invasion of Spain, there was a subtle shift in the meaning of terrorism when socialists, anarchists and communists were identified as terrorists. Despite the fact the anarchists in the nineteenth century were identified as terrorist, they were no terrorists. This is because they did not

Analyse Hitlers speech Essay Example | Topics and Well Written Essays - 1000 words

Analyse Hitlers speech - Essay Example Correctly estimating the Western Power’s reluctance to face another war, Hitler combined a series of brilliant and opportunistic strategic alliances with bold territorial acquisitions to further his aims. In 1935, Hitler began the remilitarization of Germany. This was endorsed by Great Britain, which entered into the Anglo-German Naval agreement of 1936. In the same year, Hitler formed the Axis with Italy and Japan. He exploited the resentment of the German public towards what they considered to be the unfair terms of the Treaty of Versailles to carry them along with him. A long series of provocative measures, such as Germany’s annexation of the Rhineland in March 1936, the takeover of Austria in the Anschluss of March 1938 and the annexation of the Sudetenland in September 1938, culminated in the invasion of Czechoslovakia in March 1939 (History World web site, n.d.). In the course of all these rapidly unfolding developments, Poland remained a crucial factor in Hitlerà ¢â‚¬â„¢s calculations. The document under study is an extract from the text of the speech given by Adolf Hitler to the Reichstag, or German Parliament, on 28 April 1939. Hitler’s tone regarding German-Polish relations is belligerent in the extreme. He begins by invoking the German perception of the unfairness of the Treaty of Versailles which ended World War I. According to the terms of this treaty, the German Army and Navy were limited to a token force and her Air Force was completely disbanded. Germany’s colonies were appropriated by the victorious Allies as the spoils of war. War reparations of 33 billion dollars were imposed on Germany. These terms rankled in the German psyche and were exploited by Hitler to drum up public support for his aggressive policies. A very contentious issue was the establishment of a Polish Corridor which provided Poland with access to the Baltic Sea, by linking Poland to

Zeitgeist Movie Review Example | Topics and Well Written Essays - 500 words

Zeitgeist - Movie Review Example The first part of the movie was more or less based on the birth of Jesus Chris, stating that he wasn’t infact born on this date. Here again, the director has to prove the fact that the birth date of Jesus Christ is not actually the 25th of December and that it should be in the season of winter solstice but the director has yet again avoided the theories which point otherwise. The next topic being discussed in the video is regarding the cross. The director claims that the cross is actually the symbol of the zodiac and is persistent on proving that the symbol is somewhat astrological by origin. The author has done this by discussing the myths. But it could be easily said that the cross has no relation whatsoever from astrological point of view rather it is used because Jesus Christ was crucified on the cross because crucifixion was the conventional method of punishing someone by law. Similar things can be said about the claim the author makes regarding the crown of thorns. The author also makes claim that the Bible is not actually a work of God and the stories of various prophets such as Noah, Moses mentioned in the Bible are actually again similar to other stories of the past. But the one thing that is problematic here is if the director is trying to prove wrong of the Bible why he is not trying to prove the reliability of the stories which he mentions in order to contradict the teachings of the Bible. The author also says that Horus was the God of the sun in Egypt, this shows that the author has a lack of understanding regarding the topic being discussed and he needs to do research regarding them. Every one, particularly Egyptians know that Horus was infact the God of the sky and Ra was the Sun God. This is one of the many examples which show the baseless facts that the director has mentioned in the entire video. The main conclusion that can be drawn from this video is that the religion, especially Christianity is the basis of

Set of laws governing public and private security agencies Essay

Set of laws governing public and private security agencies - Essay Example The rate of fatal crimes like homicides and cold blooded murders rose to an unacceptable level for the public. On the other hand the governments around the world blamed persistent economic recession, for their inability to invest in the betterment of the security setup. This above mentioned problem gave rise to the development of privately owned security companies, which worked for the last two decades to provide adequately acceptable security, with the help of latest technology. They are also fairly successful in fulfilling their promises, which compelled the general public to trust them more often. This trend led to the higher level of acceptability of these agencies in the society. According to some famous researchers, this fashion led to cause the shift of power needed to manage the security setup from federal agencies to general public and organizations, thus resulting in the decentralization of authority among civilians (Benson, 2007), who possess the money to hire their own ar my. This phenomenon caused the sense of inequity and insecurity among those who are weak. As a consequence, the prevalence of fear increased in the community. This practice also fueled the cold war for the authority among private and public security agencies, which impacted negatively on the standards of the already decaying security setup. However, the roots of these private security providers could be found in the large scale layoffs in military, after the end of cold war. This left, many highly skilled militants out of job. Due to this reason, they formed collations with each other and started robbing the government’s military facilities in the beginning (Schreier & Caparini, 2005). Thus with the passage of time these organizations formalized their existence and structure. Another important facet of this phenomenon is the consistent collaboration, between the government and private weapons producers, through which the former entity is getting war equipment from the latter one (Avant, 2003). This trend is playing a significant role in strengthening the weapon producers in terms of influence. These non-government security organizations are also used for the following purposes The employees of privately managed security companies are often sent on the harmony preserving missions of UN, because of the fact, that most of the developed nations do not deploy their military forces on these type of missions, due to their political reasons (Schreier & Caparini, 2005) These forces are also deployed in the case of large scale calamity effecting any nation, with the mission of helping humanity (Schreier & Caparini, 2005) They can also be used in support capacity in cooperation with other federal law enforcement agencies (Schreier & Caparini, 2005) Governing Laws for Public Reinforcement Agencies The major purpose of these organizations is to protect the citizens of the country from any kind of loss, due to theft, murder, fraud and other similar activities. They a re under strict monitoring of the government, which results in highly formalized chain of command structure (Glover, 2012). Following are the laws governing formal security operations in US implicated. The police officers cannot investigate any citizen without legitimate reasons They cannot arrest anyone without permission from government officials They cannot search anyone without

STRATEGIC RECOMMENDATION Essay Example | Topics and Well Written Essays - 1250 words

STRATEGIC RECOMMENDATION - Essay Example The corporation was initially facing stiff competition with Circuit City; however, the failure of Circuit City (a former electronics dealer) in 2009 after deep economic recession and subsequent decrease in both domestic and international demand of electronic equipments, accessories and machinery has enabled the corporation to freely play in the market and gain additional market share. Its operational stores are more than 1,000 in USA and Canada besides online presence, whereas sales in 2010 remained over $40 billions. This is an evidence of Best Buy’s financial strength, resources and abilities. In addition, the company has solid brand recognition, consumer acceptance, an established supply chain and chain network followed by a large pool of loyal (plus satisfied) consumers who have an inclination to purchase Best Buy products. Also, the consumers are inclined towards electronics and gadgets perhaps because of their fast life and heavy exposure to media that portray innovation s. It must be highlighted that Best Buys has already introduced and has adopted various customer and partner relationships improvement strategies such as focusing on after-sales services, product installation (home theatres, computer configuration etc.), product warranties, and others etc. These were not adopted in past, but have now been a major part of firm’s strategy to entice customers, improve their perceived loyalty and value, and to gain a competitive edge over its rivals something it has done better in the past than any national electronics retailer. It is also expected that it would increase the sales of company in both short and long run. However, the economic recession, reduction in households’ real incomes and purchasing because of poor employment prospects is a threat that could hinder the growth of this industry in the short run, but the recovery of economy from recession, improvement in business outlook, certainty and security in job market will improve the

Thomas Midgley Jr. Essay Example for Free

Thomas Midgley Jr. Essay Born on May 18, 1889, American engineer and chemist Thomas Midgley, Jr. was controversial for two of his discoveries. One, he discovered the Freon while working for General Motors in 1930. He intended it to be a safe non-toxic refrigerant to be used in household appliances, to replace the dangerous refrigerants used before like ammonia and propane. Freon and other CFCs soon replaced the various toxic or explosive substances previously used as refrigerants, and were later used in other applications, such as propellants in aerosol spray cans and asthma inhalers. The Society of Chemical Industry awarded Midgley the Perkin Medal in 1937 for this work. However, Midgleys discovery resulted in damage to the earths Ozone layer. Two, Midgely discovered the effectiveness of tetraethyl lead as an antiknock additive for gasoline. On October 30, 1924, Midgley participated in a press conference to demonstrate the safety of TEL. In this demonstration, he poured TEL over his hands, then placed a bottle of the chemical under his nose and inhaled its vapor for sixty seconds, declaring that he could do this every day without succumbing to any problems whatsoever. However, the State of New Jersey ordered the Bayway plant to be closed a few days later, and Jersey Standard was forbidden to manufacture TEL there again without state permission. Midgley himself was careful to avoid mentioning to the press that he required nearly a year to recover from the lead poisoning brought on by his demonstration at the press conference. He sought treatment for lead poisoning in Europe a few months after his demonstration at the press conference. Midgley was relieved of his position as vice president of GMCC but continued to work for them. Midgley died three decades before the ozone depleting effects of CFCs in the atmosphere became widely known. Another adverse effect of Midgleys work was the release of large quantities of lead into the atmosphere as a result of the large-scale combustion of leaded gasoline all over the world. High atmospheric lead levels have been associated with serious health problems. J. R. McNeill, an environmental historian, has remarked that Midgley had more impact on the atmosphere than any other single organism in Earths history.

Site Directed Mutagenesis of Subtilisin

Site Directed Mutagenesis of Subtilisin Introduction Subtilisin is a monomeric protein serine protease, which is produced by Gram positive bacteria and fungi.(1) Serine proteases are a subgroup of carbonyl hydrolase. These carbonyl hydrolases can be naturally occurring or recombinant. Naturally occurring carbonyl hydrolases consists of subtilisin matalloproteases, serine carboxypeptidase and thiol proteinase. In case of recombinant carbonyl hydrolase, the sequence coding for the wild type enzyme is altered to produce a mutant either by insertion, substitution or deletion of amino acid. Proteases are a diverse class of enzymes having several biological functions and specificities. The catalytic machinery of these enzymes is attributed to subtilisin and mammalian chymotrypsin related bacterial serine protease.(2) They are responsible for cleaving peptide bonds found in proteins. The proteases play an important role in cell wall turnover and are maximally expressed in the stationary phase. Serine proteases have molecular weights ranging f rom 25,000-30,000. They are inhibited by diidopropylfluorophosphate but are resistant to EDTA ( ethylene diamaine tetra acetic acid.) (3) The amino acid sequences of several subtilisins like subtilisin Carlsberg, subtilisin BPN, and subtilisin DY are known. Subtilisin BPN is a 275 amino acid serine protease, which is secreted by Bacillus amyloliquefaciens. This enzyme is industrially important and its gene has been cloned and expressed in Bacillus subtilis. This molecule has two enzyme binding sites- one of the site binds weakly to cations and the other one binds calcium with high affinity. These calcium binding sites are common characteristics of microbial proteases since they contribute towards kinetic and thermodynamic stability. Subtilisin is majorly used in industries wherein the environment contains high levels of metal chelators which remove the calcium from subtilisin , thus reducing its stability. It would be highly beneficial to create a stable subtilisin which would be independent of calcium. These mutated subtilisin enzymes will have an enhanced stability even in the presence of detergents and chaotropic ag ents. X-ray crystallographic analysis of mutants revealed that the conformational changes due to the mutations are localized, with very less distortion of the backbone structure. Thus a large increase in stability can be obtained with very minor changes in the protein structure. Mutants can be created by deletion, addition or substitution of amino acids at positions 75-83. The deletion of amino acids 75-83 has shown to eliminate the calcium binding site while still retaining its enzymatic activity. However, calcium free subtilisins are susceptible to proteolysis. This can be overcome by converting the active site serine 221 to cysteine (S221C). This allows the protein to fold without any auto-degradation by proteolysis. A recombinant DNA was created which encoded an active subtilisin protein without the ability to bind to calcium. The protein had no codons which specifies amino acids 75-83 and had certain additional stabilizing mutations at the N-terminal amino acids 1-8 or the ÃŽ ± helix amino acids 70-74 or the helix turn amino acids 84-89. Suitable host cells were transformed with an expression vector containing this DNA segment. Upon culturing the host cells the enzymatically active subtilisin mutant was expressed and recovered.(1) Fabric cleaning compositions containing subtilisin BPN mutants The ability to hydrolyze proteins can be exploited by incorporating these proteases as an additive to laundry detergents. Most of the stains on clothes are proteinaceous and these enzymes can efficiently aid in their removal. The amino acid sequence of the protease is responsible for its characteristics. The subtilisin BPN variants have modifications at 2-3 amino acid positions. This variant has an increased hydrolytic power and decreased adsorption to an insoluble substrate as compared to the original subtilisin. A decreased adsorption to the substrate results in a better cleaning performance. In subtilisin BPN, the amino acids ranging from position 199-220 form a large exterior loop on the molecule. This loop plays an important role in mediating the adsorption of enzyme to the surface bound peptide and mutations in this region will affect the adsorption. The substituting amino acids in case of a mutation are either neutral or negatively charged. The substituting amino acid for position 199 is mostly His, Ala, Cys, Pro, Glu, Asp, Gly, Asn or Gln and for position 207 is Glu or Asp. The cleaning compositions comprise of 0.001-1% of one or more enzyme variants. The enzyme variants can be used along with other conventional ingredients to produce efficient fabric laundry composition. These fabric cleaning compositions also contain various zwitterionic or anionic surfactants, hydrotopes, dyes or pigments, primary or secondary alcohols, chelating agents and brighteners. Fermentation: The Bacillus subtilis cells having the subtilisin mutant are allowed to grow till mid log phase in LB glucose broth and is then inoculated into a fermentor. The cells are grown overnight at 37ËÅ ¡C. Chloramphenicol is added for the antibiotic selection of mutagenized plasmid. The cells are removed from the broth by centrifugation The enzyme is purified by affinity adsorption or cation exchange chromatography. The active enzyme concentration is determined by the pNA assay. ( para nitro anilide) This assay measures the rate at which pNA is released as the enzyme hydrolyses the substrate. The rate at which the yellow colour is produced is measured at 410nm with the help of a spectrophotometer and is directly proportional to the enzyme concentration. The total protein concentration can also be estimated by determining the absorbance at 280nm. (4) Production of subtilisin variants Techniques used in microbiology, molecular biology, protein purification and recombinant DNA technology are all used in developing a subtilisin variant, which is a part of dish washing and fabric cleaning compositions. Cassette mutagenesis method for production of protease variants The gene encoding the protease is sequenced The sequence is screened for a point at which desired mutation can be made The sequences flanking this region are checked for the presence of restriction enzyme site so as to replace a short segment of the gene with an oligonucleotide which will then encode various mutants. The gene is mutated by primer extension The oligonucleotides are synthesized to have the same restriction sites, eliminating the need for synthetic linkers to create the restriction site. The genes coding for serine proteases of B.amyloliquefaciens, B.subtilis and B.licheniformis can be used as targets for mutagenesis. The gene can be inserted into a suitable vector and introduced into a host strain ( Bacillus PB92) for expression and production of mutant proteases. These mutations / substitutions enhance the performance and stability of subtilisin in detergent compositions. These serine proteases can be used in the form of granules and liquid composition both in laundry dishwashing and cosmetic applications. These enzymes are often in the form of encapsulated particles in order to protect it from the other components. Encapsulation also improves the enzyme performance and helps in regulating its availability. The encapsulating material can be derived from carbohydrates, silicates, polyvinyl alcohol, borates, PEG or paraffin waxes. Fabric cleaning performance by Blood Milk Ink microswatch assay. (BWI) This assay is performed on a microtitre plate. Samples of the subtilisin variants and reference subtilisin are obtained from filtered culture broths. 10ul samples of the enzyme are added to the BWI swatch plate along with 90ul of working detergent solution. The plates are incubated for 30 minutes. 100ul of the supernatant is transferred into a new microtiter plate and its absorbance is measured. Control wells contain the detergent solution without the protease sample. The measurement at 450nm tracks pigment removal and at 600nm tracks turbidity and cleaning. The performance index (PI) of the variant is calculated. PI compares the performance of variant and reference enzyme at the same protein concentration. PI>1 the variant is better than the wild type. PI=1 variant and standard have the same performance PI

Surgical Pain Relief: Multimodal Analgesia And Paracetamol

Surgical Pain Relief: Multimodal Analgesia And Paracetamol INTRODUCTION As a nurse on a surgical ward dealing with a diversity of operating procedures it is important to understand the effects of surgical pain, as pain intensity and control is an integral part of the nursing duties. Layzell (2008) argues that pain management for postoperative patients should be a priority for all healthcare professionals. Furthermore having a say in how medications are administered but not prescribing drugs also means that it is essential to have an understanding of pharmacology. This ensures that informed discussions can talk place with the medical officer when it is felt that pain relief for a patient requires review. Additionally this enables the right balance of pain relief to be administered to minimise distress for the patient throughout their surgical journey. According to Lucas (2008) the benefits and adverse effects of the different types of analgesia also need to be considered when treating postoperative pain. Pain is a complex phenomenon that is difficult to define. The most general definition used was published by the International Association For The Study Of Pain (IASP) in 1979 and refers to pain as an unpleasant, sensory and emotional experience arising from actual or potential tissue damage or described in terms of such damage (IASP  2010). Pain is also subjective and the intensity can only be experienced by the sufferer (Mann and Carr 2006). Neurophysiology there are three types of pain receptors communicating pain signals through nociceptors or pain fibres (Mann and Carr 2006). These receptors can be found in the skin, surfaces of the joints, periosteum (the specialised lining around the bone), arterial walls and certain structures in the skull, although the brain itself does not have any of these receptors (Mann and Carr 2006, p3). Each receptor reacts to a different stimulus. The mechanical receptor to touch, thermal to heat or cold and chemical to products present in the body that are released after trauma causing inflammation and increased sensitivity at the wound site (Mann and Carr 2006). The chemical receptors also react to chemicals introduced into the body (Mann and Carr 2006). The nociceptors are the sensory instruments that transmit pain signals through sensory nerve fibres to the dorsal horn of the spinal cord and into the brain (Mann and Carr 2006). There are three types of nerve fibres: A-delta responds to mechanical or thermal sensations, C fibres also known as polymodal because they respond to mechanical, thermal and chemical influences and Aà ¢Ã¢â€š ¬Ã¢â‚¬Ëœbeta fibres which occur in the skin, reacts to touch but does not transmit pain sensations (Mann and Carr 2006). Psychologically pain can induce fear, anxiety, anger, frustration and also a sense of helplessness may be experience from being unable to physically control the intensity of the pain being felt (Rothrock et. al. 2007). Physiologically pain can reduce the functions of the immune system whilst increasing the potential for wound and chest infections as well as impairing the wound healing processes (Middleton 2003, Pudner and Ramsden 2010). Pain also induces vomiting, increases the workload of the cardiovascular and gastrointestinal systems, decreases lung capacity and can also reduce physical mobility (Middleton 2003, Rothrock et. al. 2007). Traditionally following surgery, a single opioid drug such as morphine is used, depending on the type of surgery performed, for moderate to severe or acute pain (Shorten et. al. 2006). It is argued that patients who receive this monoà ¢Ã¢â€š ¬Ã¢â‚¬Ëœtherapy would prefer to be treated with non Ãƒ ¢Ã¢â€š ¬Ã¢â‚¬Ëœopioid remedies (Shorten et. al. 2006). According to Mann and Carr (2006) using the monoà ¢Ã¢â€š ¬Ã¢â‚¬Ëœtherapy method only targets one pain pathway and although morphine is considered the gold standard it tends to have many adverse effects. These include a 0.2% risk of respiratory depression, sedation, urinary retention, nausea and vomiting which affects around 30% of patients, itching or pruritus, hypotension or low blood pressure plus confusion and hallucinations in the elderly (Mann and Carr 2006, Rothrock et. al. 2007, Manley and Bellman 1999). Some of these side effects are controlled with antià ¢Ã¢â€š ¬Ã¢â‚¬Ëœemetics for sickness and antià ¢Ã¢â€š ¬Ã¢â‚¬Ëœhistamines for pruritus (Rothrock et. al. 2007). Combinational drug therapy began in the 1950s (Michielsen 2007). Since then there has been increasing developments in establishing opiate sparing analgesic regimes with fewer side effects for surgical pain (Shorten et. al. 2006). Painkillers such as paracetamol (acetaminophen) can be combined with drugs from the codeine group, tramadol and non-steroidal antià ¢Ã¢â€š ¬Ã¢â‚¬Ëœinflammatory drugs (NSAIDs) as part of a multimodal regime (Manley and Bellman 1999). This allows for lower doses of individual drugs to be given, reducing the severity of adverse events and targeting more than one pain pathway as each drug has a different mechanism of action (Shorten et. al. 2006). Paracetamol is a universal drug that is generally well tolerated but its mechanisms are not fully understood (Mann and Carr 2006). There are only mild to rare reported occurrences of side effects such as skin rashes and other allergic reactions (Manley and Bellman 1999). Paracetamol, a member of the non-opioid group is one of oldest known synthetic analgesic and antipyretic drugs (Manley and Bellman 1999, p470). Being also an antipyretic paracetamol has the ability to reduce fever temperature (Pudner and Ramsden 2010). It can be administered orally, rectally or intravenously in the form of a prodrug known as perfalgan or propacetamol (Manley and Bellman 1999, Royal Pharmaceutical Society of Great Britain 2007). Prodrugs are treatments that need to be broken down in the body before they become active (MedicineNet 2010). It is suggested that paracetamol should be used as part of a multimodal regime for surgical patients experiencing mild to moderate pain (Pudner and Ramsden 2010). Multimodal treatments involve combining drugs to form a compound in order to increase pain relief and reduce opioid adverse effect (Shorten et. al 2006, Manley and Bellman 1999, Pudner and Ramsden 2010). These combinational drugs are regulated by the Medicines and Healthcare Products Regulatory Agency (MHRA) and European Medicines Evaluation Agency (EMEA) and only a limited number has been approved (Shorten et. al 2006, Department of Health 2010). Pharmaceutical companies have also introduced several fixedà ¢Ã¢â€š ¬Ã¢â‚¬Ëœdoseà ¢Ã¢â€š ¬Ã¢â‚¬Ëœcombinations such as coà ¢Ã¢â€š ¬Ã¢â‚¬Ëœcodamol a combination of codeine phosphate, a weak opioid and paracetamol as well as coà ¢Ã¢â€š ¬Ã¢â‚¬Ëœdydramol a compound of dihydrocodeine and paracetamol (Shorten et. al 2006, p185). The main side effect of these codeine products is constipation, which can be remedied with a mild laxative (Manley and Bellman 1999). Tramadol, another weak opioid can also be combined with paracetamol (Manley and Bellman 1999). The side effects of tramadol include minimal respiratory depression, nausea, vomiting, dizziness, headache and sweating (Manley and Bellman 1999). Using this drug appears to defeat the object of reducing opiate contraindications but the combination is generally well tolerated and effective for moderate to severe pain (Shorten et. al. 2006). Rothrock et. al (2007) argues that combining NSAIDs with opioid drugs effectively reduces opioid usage by 20-40%. However increased postoperative bleeding and the chances of developing gastrointestinal ulcers cause NSAIDs to be used sparingly for surgical patients ((Rothrock et. al. 2007). The Human Rights Act 1998 states that ethically it is the duty of all healthcare staff to ensure that patients are protected from any form of torture, inhuman and degrading treatment and punishment (Office of Public Sector Information 1998, Article  3). Nurses are also responsible for their actions and as such must be able to justify decisions made or omissions which affect the wellà ¢Ã¢â€š ¬Ã¢â‚¬Ëœbeing of a patient (Nursing and Midwifery Council 2008). From a surgical nursing perspective this means it is imperative to ensure patients receive good pain management following their operation. However according to a recent study by Dolin, Cashman and Bland (2002) one in five patients still report severe postoperative pain. Manley and Bellman (1999) suggest patients commonly believe that pain is acceptable following surgery. While Pudner and Ramsden (2010) argue that postoperative pain should be controlled and patients should not expect or see pain as inevitable. This literature review aims to firstly evaluate current research and evidence in relation to the use of paracetamol as one part of a multimodal analgesia regime for surgical pain relief. Secondly to use the results to make recommendations for standardising multimodal pain control for postoperative patients and re-educate staff on the importance of effective pain management. METHODOLOGY Search Criteria For this literature review an advanced search was carried out over the internet. The health and medical sciences specific databases of Cumulative Index to Nursing and Allied Health Literature (CINAHL), Medical Literature Online (MEDLINE), Cochrane Library and Internurse.com were explored (Thomas 2000). CINAHL includes full text and is the most relevant source of information for nursing while MEDLINE focuses on life sciences and is produced by the National Library of Medicine (LoBiondo-Wood 2010, p68). The Cochrane Library holds a collection of systematic reviews and Internurse.com has journal articles written by nurses (LoBiondo-Wood 2010, p68). The Cochrane Library was used to determine if any critical reviews had previously been carried out on the subject of paracetamol being used as part of a multimodal regime for postoperative patients. Only 4 papers were found. Internurse.com was exploited for articles containing current knowledge on the use of multimodal therapy for surgical pa tients. These editorials were utilised within the introduction. The keywords or inclusion criteria for the literature search included paracetamol in the title while the words surgical and post operative were left optional to increase the depth of the search. Truncation or wildcards were employed to make the search more sensitive and specific to the topic being researched as follows: surgical surg* and post operative post op* (Gerrish, and Lacey 2006). The search mode was set to Boolean which defines the relationships between words or groups of words in a literature search (LoBiondo-Wood 2010). This process involved using the word AND before the truncated words surg* and post op*. The date time frame was also limited from 2000 to 2010 to ensure that the studies were up to date and relevant to present day policies and procedures for pain control (LoBiondo-Wood 2010). Restrictions were also placed to only include papers that were based on humans, research papers and in the English language. Humans were selected as experiments on animals due to their biological makeup was not considered to be relevant to controlling postoperative pain in human beings. As this is a literature review it was appropriate to only select research papers for analysis. Language was also deemed to be significant as finances and the time schedule to complete the review did not allow for interpretation of the papers from other Dialects. Expanders were included to find papers that had related words and for the search to be carried out within the full text of the articles. Review 8 papers resulted from the above search criteria. These were then screened using inclusion criteria, the titles and abstracts to determine their relevance to relieving surgical pain with multimodal analgesia therapy. The inclusion criteria was trials that included patients who had received paracetamol (acetaminophen) postoperatively, trials that included multimodal therapy, papers that were published within the last 10 years, subjects who were adults as my surgical setting only treats patients over the age of 18 and within a hospital environment. Exclusion criteria were trials that involved animals or children as discussed earlier. The types of interventions could include any routes for drug administration as paracetamol can be administered via intravenous, oral or rectal modes. One paper was excluded from the review at this point as it was a monoà ¢Ã¢â€š ¬Ã¢â‚¬Ëœtherapy trial for paracetamol. The remaining 7 papers met with the inclusion criteria and were scored for methodological quality using a critical appraisal skills programme (CASP) containing 10 questions {{488 Public Health Resource Unit (PHRU) 2007}}. The questions were answered yes, no or cant tell for each paper. Using a tool provides a way of systematically appraising what is published and filtering through papers to determine their relevance and accuracy {{427 Crookes, P. and Davies, S. 2004}}.

SECURITY :: essays research papers

A firewall is a set of related programs, located at a network gateway server, that protects the resources of a private network from users from other networks. (The term also implies the security policy that is used with the programs.) An enterprise with an intranet that allows its workers access to the wider Internet installs a firewall to prevent outsiders from accessing its own private data resources and for controlling what outside resources its own users have access to. Basically, a firewall, working closely with a router program, examines each network packet to determine whether to forward it toward its destination. A firewall also includes or works with a proxy server that makes network requests on behalf of workstation users. A firewall is often installed in a specially designated computer separate from the rest of the network so that no incoming request can get directly at private network resources. There are a number of firewall screening methods. A simple one is to screen requests to make sure they come from acceptable (previously identified) domain name and Internet Protocol addresses. For mobile users, firewalls allow remote access in to the private network by the use of secure logon procedures and authentication certificates. A number of companies make firewall products. Features include logging and reporting, automatic alarms at given thresholds of attack, and a graphical user interface for controlling the firewall. Encryption is the conversion of data into a form, called a ciphertext, that cannot be easily understood by unauthorized people. Decryption is the process of converting encrypted data back into its original form, so it can be understood. The use of encryption/decryption is as old as the art of communication. In wartime, a cipher, often incorrectly called a "code," can be employed to keep the enemy from obtaining the contents of transmissions. (Technically, a code is a means of representing a signal without the intent of keeping it secret; examples are Morse code and ASCII.) Simple ciphers include the substitution of letters for numbers, the rotation of letters in the alphabet, and the "scrambling" of voice signals by inverting the sideband frequencies. More complex ciphers work according to sophisticated computer algorithms that rearrange the data bits in digital signals. In order to easily recover the contents of an encrypted signal, the correct decryption key is required. The key is an algorithm that "undoes" the work of the encryption algorithm. Alternatively, a computer can be used in an attempt to "break" the cipher.

Xena vs. The Odyssey :: Compare Comparing Contrast

Xena vs. The Odyssey In this episode of "Xena: The Warrior princess," the story of Odysseus return to Ithaca after the Trojan War is told. The episode starts when Xena and Gabriel are walking down a beach and they see a man fighting alone against a small army, so they decide to help him out. After the battle was over, they found out that the man fighting was Odysseus. He told Xena his story and how he had been traveling for ten years to get home because Poseidon was angry with him. Xena and Gabriel decide to help him get to Ithaca, so they sail off. First they got cut up in a terrible storm that Poseidon sent them. Most Odysseus' men(of the ones he had left after ten years) died here, but still the ship survived. After this they had to sail close to the island of the sirens. Xena knowing the effect of their singing in men tied up every man left in the boat including Odysseus. The closed they got to the island the stronger the singing. The effect of the voices of the sirens was so strong that Odysse us almost broke away to go to them, but then Xena started singing really loud to contra rest the effect. Finally Xena wan they sail safely through the island. Finally they got to Ithaca, where Odysseus found out that Penelope's hand would be given in marriage to the winner of the contest taking place the next day. Odysseus, helped by Xena, also found out that Penelope had been faithful to him all those years by rejecting every suitor she had. Yet after ten years, they had finally pressure her to decide, so she decided that whomever wan the contest would be her husband. The contest consisted of the trial of the bow, every suitor had to first string it and then shut an arrow through 12 arrows. None even got close, until Odysseus tried it, he was really close to making it. However, he could not quite do it alone, so Xena without anyone seen her, nor even Odysseus himself, help him out. Ones Odysseus had wan the contest he revealed his identity and helped by Xena, they killed all of the suitors. During the trip, Xena and Odysseus got quite close, in fact they felt in love, so when Odysseus attended to leave his kingdom to Penelope to go with Xena, she convinced him that it was not wise and that his place was with his wife, who had been waiting for him for ten years, ruling Ithaca.

Despite the fact that Internet Explorer 5. 5 is one of the most popular in companies, I’d like to recommend another one – Maxthon Browser 2. 0, because it is much more convenient for office working. Maxthon Internet Browser 2. 0 is a powerful tabbed browser nowadays. Its main advantage is a highly customizable interface. Although Maxthon Browser is based on the engine of Internet Explorer, it has many additional efficient features. What works in Internet Explorer works as well in Maxthon Browser and even better and faster. (Maxthon). Maxthon Browser includes many new features involving full customization of layout, new clean interface, multi-tab workspace, better security and improved rss/podcast reader. Until nowadays a big inconvenient was: bookmarklet, toolbar and 3rd party plugins. Maxthon 2. 0 supports all of the functions compared with Internet Explorer, Opera, etc. Additionally Maxthon has few exclusive screenshots below. Nevertheless the most interesting feature of new Maxthon is its possibility to create an account and to store all necessary information and settings online. The function is simply used just by logging in. Although Opera, Internet Explorer and FireFox are more sophisticated and have got more market share, Maxthon is swiftly gaining popularity. (Maxthon) Today is the era of Internet and application functionality is able to expand beyond the operating system into the desktops and the web. Personalization and delivery is very important and Maxthon Browser with its organic growth is a serious representative of such trend. In several years Maxthon will become a key player in the Internet market, because it has serious investors such as Morten Lund, CRV, and WI Harper, growing community and actually beautiful product. I recommend Maxthon, because it differs from other browsers and makes working process with Internet easier as it includes the visible gestures and customizable Tab handling. But the most useful is if the browser is closed accidentally with ten tabs open, the next time it is opened, they are still there. (Maxthon). References â€Å"Maxthon: the Browser that Rocks†. Argumentative Essay on Teen Pregnancy

immature motherliness is a major hump in the United States. Some large number say that this is societys fault. In conclusion, I believe that teenage pregnancy is a actually important issue. The sexual society of todays America has resulted in a large increase in teen pregnancy. Mansarays and Breelers stories could be apply at bottom the kinfolk program to stress several cultivation points, specially those extend to to reinforcing the childs federal agency and social awareness. In that way, these resources nates rattling be used to support childrens study and development. The authors may actually discover extending their work into other literature by expanding these stories into serial publication that semi different issues and competitivenesss children administration from day to day.A good way to go across these resources in the schoolroom setting is to leave alone each student read, essay and get word these stories in their protest ways. This means that what a child learns socially is connect to what he/she learns cognitively, and ungodliness versa. What a child learns by interacting with other large number adds to his/her own precis and indeed increases his/her consciousness in some cognitive processes.
